Is the ESP32-S3-DevKitC-1 compatible with 5V sensors?

Not directly. As with the entire ESP32 family, the logic voltage on the pins is strictly 3.3V. Connecting a 5V signal directly to a GPIO will fry the microcontroller. You must always use a Level Shifter to read or send signals to 5V modules.

What drivers do I need and which USB port should I use?

For initial programming (flashing), it is recommended to use the UART port (usually labeled COM/UART) which requires the drivers for the onboard Communication Chip (e.g., CH343/CP2102). The second USB port provides Native communication (JTAG/OTG) and is recognized automatically as a USB device by the operating system.
